La glucosa-6-fosfato deshidrogenasa (G6PDH) como blanco molecular contra Trypanosoma cruzi
Resumen:
La enfermedad de Chagas es la manifestación de la infección sistémica y crónica causada por el parásito Trypanosoma cruzi, actualmente considerada endémica en América. El tratamiento actual se basa en dos fármacos: Nifurtimox y Benznidazol. Ambos presentan una elevada toxicidad produciendo efectos secundarios severos, siendo un tratamiento complicado y poco accesible, ya que requiere de tiempos prolongados y altos costos. Por otra parte, tampoco han demostrado buena eficacia en la fase crónica y se ha evidenciado la aparición de resistencia a ambos medicamentos. Por estos motivos es de gran importancia contribuir al desarrollo de nuevos fármacos para controlar esta “enfermedad desatendida”. En este trabajo se plantea como objetivo estudiar a la enzima glucosa-6-fosfato deshidrogenasa (G6PDH) como blanco molecular de T. cruzi, ya que ésta ha mostrado, en diversos estudios, ser un factor de virulencia para las formas infectivas. Esta proteína es responsable de catalizar la primera reacción de la vía de las pentosas fosfato, generando NADPH, ribosa-5-fosfato e intermediarios de la glicólisis. Por tanto, las moléculas capaces de inhibir esta enzima crucial para el metabolismo del parásito constituyen potenciales fármacos a explorar para el tratamiento de la enfermedad de Chagas.
Chagas disease is a systemic and chronic infection caused by the parasite Trypanosoma cruzi, currently considered endemic in America. The current treatment is based on two drugs: Nifurtimox and Benznidazol. Both of them present high toxicity, producing severe side effects, being a complicated and little accessible treatment, since it is long and expensive. Current drugs have not demonstrated good efficacy in the chronic phase either, and the emergence of resistance to both of them has been reported. For these reasons, it is of great importance to contribute to the development of new drugs to control this “neglected disease.” In this work, the enzyme glucose-6-phosphate dehydrogenase (G6PDH) was tested as a molecular target of T. cruzi, since it was shown to be a virulence factor for the infective forms. This protein catalyzes the first reaction of the pentose phosphate pathway, generating NADPH, ribose-5-phosphate and glycolysis intermediates. Therefore, the molecules capable of inhibiting this crucial enzyme constitute potential drugs for the treatment of Chagas disease.
